Abstract:
Antarctickrill(Euphausiasuperba)isavaluablemarineresourceknownforitsrichnutritionalandfunctionalproperties.ThisstudyaimedtooptimizethepreparationprocessofAntarctickrillpeptideandanalyzeitsα-glucosidaseinhibitoryactivityusingresponsesurfacemethodology(RSM).Theresultsdemonstratedthesignificanteffectofprocessvariablesontheyieldandbioactivityofthepeptide.
Introduction:
Antarctickrillisaspeciesofshrimp-likecrustaceanthatthrivesinthecoldwatersnearAntarctica.Itisarichsourceofprotein,lipids,andotherbeneficialcompounds,includingpeptideswithvariousbioactivities.Inrecentyears,researchonthepotentialhealthbenefitsofAntarctickrillpeptideshasgainedsignificantattention.
Methods:
1.RawMaterialPreparation:Antarctickrillisharvestedandimmediatelyprocessedtoretainitsfreshnessandnutritionalvalue.Thekrillisthendriedandgroundintoafinepowdertofacilitatesubsequentprocessing.
2.ExtractionofPeptides:TheAntarctickrillpowderissubjectedtoenzymatichydrolysistoreleasethepeptides.VariousenzymessuchasAlcalase,Flavorzyme,andNeutrasecanbeusedforhydrolysis.
3.OptimizationofProcessVariables:TheRSMisemployedtooptimizetheprocessvariablesincludingenzymeconcentration,temperature,pH,andhydrolysistime.Acentralcompositedesign(CCD)isusedtodesigntheexperiments,andthedataobtainedisanalyzedusingregressionanalysistobuildmathematicalmodels.
4.DeterminationofPeptideYield:Thepeptideyieldisdeterminedbymeasuringtheconcentrationofpeptidesusinganappropriateanalyticaltechnique,suchastheBradfordassayorhigh-performanceliquidchromatography(HPLC).
5.α-GlucosidaseInhibitoryActivityAnalysis:TheinhibitoryactivityoftheAntarctickrillpeptideonα-glucosidaseisassessedusinganinvitroassay.Theenzymeactivityismeasuredbymonitoringthehydrolysisofsubstrate,andtheinhibitorypotencyisexpressedasIC50value(concentrationrequiredtoinhibit50%ofenzymeactivity).
