Abstract:
Estrogensareagroupofimportanthormonesthatplayvitalrolesintheregulationofvariousphysiologicalprocessesinanimals.Thepresenceofestrogensintheenvironmentposespotentialriskstobothwildlifeandhumanhealth.Therefore,developinganalyticalmethodsfortheaccuratedeterminationofestrogensinbiologicalsamplesiscrucial.Inthisstudy,amethodbasedonsolid-phaseextraction(SPE)coupledwithultra-high-performanceliquidchromatography-tandemmassspectrometry(UHPLC-MS/MS)wasdevelopedforthesimultaneousdeterminationofnineestrogensinbullfrogs.
Introduction:
Estrogensaresteroidhormonesthatareprimarilyresponsibleforthedevelopmentandmaintenanceoffemalecharacteristicsinvertebrates.However,theyarealsopresentinmales,albeitatlowerconcentrations.Estrogenshavebeenwidelydetectedintheenvironment,mainlyasaresultofhumanactivitiessuchasthereleaseofpharmaceuticals,householdproducts,andindustrialwaste.Duetotheirendocrine-disruptingeffects,estrogenshaveattractedsignificantattentioninrecentyears.
Methodology:
1.SamplePreparation:
-Bullfrogsampleswerecollectedandhomogenized.
-Thehomogenizedsamplesweresubjectedtosolid-phaseextraction(SPE)usingC18cartridgestoextractthetargetestrogens.
-TheeluatefromtheSPEwascollectedandevaporatedtodrynessunderreducedpressure.
-TheresiduewasreconstitutedwithanappropriatesolventforUHPLC-MS/MSanalysis.
2.Instrumentation:
-TheUHPLC-MS/MSsystemconsistedofanultra-high-performanceliquidchromatographysystemcoupledtoatandemmassspectrometer.
-Theseparationofestrogenswasachievedusingareverse-phaseC18column.
-Themassspectrometerwasoperatedinmultiplereactionmonitoring(MRM)mode.
3.MethodValidation:
-Thedevelopedmethodwasvalidatedintermsoflinearity,sensitivity,accuracy,precision,recovery,andmatrixeffects.
-Calibrationcurveswereconstructedbyanalyzingstandardsolutionsofestrogensatvariousconcentrations.
-Thelimitso
