Abstract:
Thegeneticdiversityofaplantpopulationisafundamentalaspectofitsevolutionarypotentialandecologicaladaptability.Inthisstudy,weinvestigatedthegeneticdiversityoftheRedChinaberry(Meliaazedarach)populationthroughtheuseofSimpleSequenceRepeat(SSR)markers.SSRanalysisprovidedimportantinsightsintothegeneticstructure,geneflow,andevolutionaryhistoryofthespecies.OurresultshighlightedthehighgeneticdiversityandpotentialforadaptationwithintheRedChinaberrypopulation,emphasizingtheimportanceofconservationeffortstopreserveitsgeneticresources.
Introduction:
TheRedChinaberry(Meliaazedarach)isawidelydistributedtreespecieswitharichhistoryoftraditionalmedicinalandculturaluses.Thespeciesalsoexhibitsvariousecologicalroles,suchasprovidingshade,timber,andshelterforbirdsandinsectsalike.StudyingthegeneticdiversityofRedChinaberryiscrucialforunderstandingitsevolutionarypotential,populationdynamics,anddevisingeffectiveconservationstrategies.SSRmarkersarewidelyusedinpopulationgeneticsstudiesduetotheirco-dominantinheritance,highpolymorphismlevels,andeaseofdetection.
Methods:
Atotalof100RedChinaberryindividualsweresampledfromdifferentlocationsacrossitsdistributionrange.GenomicDNAwasextractedfromeachindividualandPCRamplificationwasperformedusingasetofselectedSSRmarkers.Theamplifiedproductswereseparatedonpolyacrylamidegelsandvisualizedusingstandardsilverstainingmethods.TheresultingSSRprofileswerescored,andgeneticdiversityparameterssuchasthenumberofalleles,allelicrichness,andgenediversitywerecalculated.Geneticstructureanalysiswasconductedtoassessthepopulationdifferentiation,geneflow,andgeneticclusteringusingsoftwareprograms.
Results:
OurSSRanalysisrevealedhighlevelsofgeneticdiversitywithintheRedChinaberrypopulation.AtotalofXallelesweredetected,withanaverageofXallelesperlocus.TheobservedandexpectedheterozygosityvalueswereXandX,respectively,indicatingahighlevelofgeneticvariability.Theallelicri
