Introduction:
Understandingthemolecularmechanismsunderlyingmuscledevelopmentandintramuscularfat(IMF)depositioninchickensfromtheembryonictotheearlygrowthstageisofgreatimportanceforimprovingmeatqualityandproductionefficiency.ThisresearchaimstoinvestigatetheproteinprofilesassociatedwithmuscledevelopmentandIMFdepositionduringthiscriticalperiod.Thefindingswillprovideinsightsintotheregulatorymechanismsandpotentialtargetsforenhancingmeatqualityinpoultryproduction.
Objective:
ThemainobjectiveofthisresearchistoanalyzetheproteincompositionandexpressionpatternsassociatedwithmuscledevelopmentandIMFdepositioninchickensfromtheembryonicstagetoearlygrowthstage.Thespecificgoalsinclude:
1.CharacterizethedynamicchangesinproteinexpressionprofilesduringmuscledevelopmentandIMFdeposition.
2.IdentifykeyregulatoryproteinsandpathwaysinvolvedinmuscledevelopmentandIMFdeposition.
3.Investigatetherelationshipbetweendifferentiallyexpressedproteinsandmeatqualitytraits.
Methods:
1.SampleCollection:
-Collectchickenmusclesamplesatvariousdevelopmentaltimepoints,includingembryonicstages(E10,E14,E18),andearlygrowthstages(1week,3weeks,6weeks).
-Obtainmusclesamplesfromatleast20chickenspertimepointtoensurestatisticalsignificance.
2.ProteinExtraction:
-Homogenizemusclesamplestoextracttotalproteinsusingastandardprotocol.
-QuantifyproteinconcentrationusingaBicinchoninicAcid(BCA)assay.
3.Two-DimensionalGelElectrophoresis(2-DE):
