Abstract:
Cytoplasmicmalesterility(CMS)isacommonphenomenoninplantspecies,includingwheat.UnderstandingthegeneticbasisofCMSisessentialfordevelopinghybridwheatvarieties.Inthisstudy,weemployedtheRandomAmplifiedPolymorphicDNA(RAPD)techniquetoanalyzetheG-typewheatCMSline.TheobjectivewastoidentifymolecularmarkerslinkedtoCMSandgaininsightsintotheunderlyinggeneticmechanisms.OurfindingsshedlightonthepotentialapplicationsofmolecularmarkersforCMSpredictionandthebreedingofhybridwheatvarieties.
Introduction:
Cytoplasmicmalesterility(CMS)isanimportanttraitinplantbreeding,particularlyforhybridseedproduction.InG-typewheat,CMSisprimarilycontrolledbycytoplasmicmitochondrialgenes.TheidentificationofmolecularmarkerslinkedtoCMSgenescanfacilitatethedevelopmentofmolecularbreedingstrategiesforproducinghybridwheatvarieties.RAPDtechniquehasbeenwidelyusedinmolecularmarkeranalysisduetoitssimplicityandefficiency.Inthisstudy,weemployedRAPDmarkerstoinvestigatetheinheritanceofcytoplasmicmalesterilityinG-typewheat.
Methods:
1.Plantmaterials:TheG-typewheatCMSlineanditscorrespondingmaintainerlinewereusedinthisstudy.
2.DNAextraction:GenomicDNAwasextractedfromyoungleavesusingacommercialDNAextractionkit.
3.RAPDanalysis:RAPDprimerswererandomlyselectedfromapoolofprimersandusedtoamplifyDNAfragments.TheamplifiedfragmentswereseparatedbyagarosegelelectrophoresisandvisualizedunderUVlight.
4.Dataanalysis:RAPDbandswerescoredandanalyzedusingappropriatestatisticalmethods.SimilaritymatriceswereconstructedusingJaccard'scoefficient,andclusteranalysiswasperformedtoidentifygeneticrelationships.
Results:
1.IdentificationofCMS-associatedRAPDmarkers:SeveralRAPDmarkerswerefoundtobeassociatedwiththeCMStraitinG-typewheat.ThesemarkersexhibitedpolymorphicbandpatternsbetweentheCMSlineandthemaintainerline.
2.Clusteranalysis:ThegeneticrelationshipamongdifferentwheatlineswasanalyzedusingRAPDmarkers.TheCMSlineformedadistinctcluster,indicatingge
