小麦胚性非再生细胞中游离氨基酸分析 Title:AnalysisofFreeAminoAcidsinWheatEmbryogenicNon-regenerativeCells Abstract: Wheatembryogenicnon-regenerativecellsholdgreatpotentialforplanttissuecultureandcropimprovement.Understandingtheircellularcomposition,specificallytheprofileoffreeaminoacids,iscrucialforoptimizingthetissuecultureconditionsandenhancingtheregenerationabilityofembryogeniccells.Inthisstudy,weperformedananalysisoffreeaminoacidsinwheatembryogenicnon-regenerativecellsanddiscusstheirbiologicalsignificance. 1.Introduction: Wheatembryogenicnon-regenerativecellsarecapableofformingembryogeniccallusbutfailtoregenerateintoplantlets.Theunderlyingreasonsfortheinabilitytoregeneratearemultifaceted,andmetabolicalterationsaresuggestedtoplayacrucialrole.Aminoacidsarekeymetabolitesinvolvedinvariouscellularprocesses,includingproteinsynthesis,stressresponse,andhormonesignaling.Thus,analyzingthefreeaminoacidprofilecanprovideinsightsintothephysiologicalandmolecularmechanismsunderlyingtheregenerativeabilityofwheatembryogeniccells. 2.Methods: 2.1SamplePreparation:Wheatembryogenicnon-regenerativecellswereculturedonasuitablemedium,andafterreachingthedesiredgrowthstage,cellswereharvested,frozen,andlyophilized.Thelyophilizedcellswerepulverizedtoafinepowderforfurtheranalysis. 2.2AminoAcidExtraction:Thepulverizedcellswereextractedusinganappropriatesolvent,suchas80%coldmethanol,toobtainasamplecontainingfreeaminoacids. 2.3AminoAcidAnalysis:Theextractedsamplewassubjectedtoaminoacidanalysisusinghigh-performanceliquidchromatography(HPLC).TheHPLCsystemequippedwithasuitableaminoacidanalyzercolumnandaUVdetectorenabledtheseparationandquantificationofindividualaminoacids. 3.Results: Theanalysisoffreeaminoacidsrevealedadiversespectrumofaminoacidspresentinwheatembryogenicnon-regenerativecells.Themajoraminoacidsdetectedincludeglutamicacid,asparticacid,proline,serine,andglycine.Additionally,essentialaminoacids,suchasleucine,lysine,andphenylalanine,werealsopresentinsignificantquantities.Notably,someaminoacidsshowedmarke
