Introduction:
Purplelaver(Porphyraspp.)isoneofthemostimportantseaweedsconsumedbyhumans.Itisrichinprotein,vitamins,andmineralsandisusedinmanytraditionalcuisinesofAsiancountries.InChina,Japan,andKorea,cultivationofpurplelaverhasbeenpracticedforthousandsofyears.Today,thereisanincreasingdemandforpurplelaverduetoitsnutritionalvalueandmedicinalproperties.However,thequalityandyieldofpurplelavervaryamongdifferentcultivarsorstrains.Therefore,itisimportanttounderstandthediversityandgeneticrelationshipsamongpurplelavercultivarsinordertoimprovetheirproductionandquality.
ISSRanalysis:
Theinter-simplesequencerepeat(ISSR)techniquehasbeenwidelyusedingeneticdiversityanalysisofvariousplantspecies.ItisaPCR-basedDNAfingerprintingmethodthatamplifiesinter-simplesequencerepeatregionsbetweentwoarbitraryprimers.Inthisstudy,ISSRanalysiswasperformedonfivedifferentpurplelavercultivars,including:
1.T7402
2.T7405
3.T7412
4.T7507
5.T7509
Atotalof12ISSRprimerswereusedtoamplifythegenomicDNAofpurplelavercultivars.Theamplifiedproductswereseparatedbyelectrophoresison1.5%agarosegelsandvisualizedusingethidiumbromidestaining.Thebandingpatternswerescoredforeachcultivar,andabinarymatrixwasconstructedfordataanalysis.
Results:
TheISSRanalysisproducedatotalof115clearandreproduciblebands,rangingfrom150to2500bpinsize.Amongthem,97bandswerepolymorphic,andthepercentageofpolymorphicbands(PPB)was84.4%.Thenumberofbandsperprimerrangedfrom6to16,withanaverageof9.6bandsperprimer.ThehighestnumberofbandswasobtainedbyprimerISSR-5,whilethelowestnumberofbandswasobtainedbyprimerISSR-2.
ThegeneticsimilaritybetweencultivarswascalculatedbasedontheJaccardcoefficient,andadendrogramwasconstructedusingtheUPGMAmethod.Thedendrogramshowedthatthefivecultivarscouldbeclassifiedintotwogroups.GroupIconsistedofT7402,T7405,andT7412,whilegroupIIconsistedofT7507andT7509.ThegeneticdistancebetweengroupIandgroupIIwas0.54,indicatingsignificantgeneticdifferentiationbetweenthetwogroups.
