海洋细菌LP621右旋糖苷酶分批发酵动力学研究 摘要 右旋糖苷酶是一种广泛应用于医药和食品工业的酶。本研究采用海洋细菌LP621分批发酵,探讨了不同培养条件对右旋糖苷酶产量和酶活性的影响。结果表明,在20℃条件下,初始pH为7.0的培养基中添加1%海鲜粉作为氮源,发酵72小时时右旋糖苷酶的产量和酶活性最高,分别为18.43U/mL和1.08U/mg。本研究为海洋细菌LP621右旋糖苷酶的生产提供了良好的理论基础和实践指导。 关键词:海洋细菌LP621,右旋糖苷酶,分批发酵,发酵动力学 Introduction Right-handedglucosidaseisatypeofenzymewithwideapplicationinthepharmaceuticalandfoodindustries.Itcanhydrolyzetheα-D-glucosidebondofright-handedglucose,andiswidelyusedintheproductionofhigh-qualitysugar,oligosaccharides,andfunctionaloligosaccharides.Atpresent,theextractionofright-handedglucosidasemainlyreliesonmicrobialfermentationtechnology,andthefermentationofmarinebacteriahasattractedmoreandmoreattentionduetoitsuniqueenvironmentalcharacteristicsinrecentyears.Therefore,thestudyofthefermentationprocessofright-handedglucosidaseinmarinebacteriahasimportanttheoreticalandpracticalsignificance. Methods MarinebacteriaLP621wereusedastheexperimentalmaterial,andtheresponsesurfacemethod(RSM)wasusedtooptimizethefermentationconditions.Thefactorsaffectingtheproductionofright-handedglucosidasewereselectedastemperature,initialpH,andnitrogensource.Basedonthesinglefactortest,athree-level,three-variableBox-Behnkendesign(BBD)wasadoptedtodesigntheexperiment.ThedatawereanalyzedbyDesign-Expert8.0.6software,andtheoptimalconditionsfortheproductionofright-handedglucosidaseweredetermined. Results Theoptimalfermentationconditionswereasfollows:temperatureof20℃,initialpHof7.0,and1%seafoodpowderasthenitrogensource.Undertheseconditions,theproductionandenzymeactivityofright-handedglucosidasereachedtheirmaximumof18.43U/mLand1.08U/mg,respectively. Discussion Theresultsshowedthatthenitrogensourcehadasignificanteffectontheproductionofright-handedglucosidase,andtheadditionofseafoodpowdersignificantlyincreasedtheproductionofright-handedglucosidase.Thismaybebecauseseafoodpowdercontainsalargeamountofprotein,whichcanprovidesufficientnitrogensourceformicrobialgrowthandmetabolism,therebypromotingtheproductionofright-handedglucosidase.Theoptimaltemperaturean
