Abstract:
Purinesareimportantnitrogenouscompoundspresentinvariousfoodsandbeverages.Theexcessiveintakeofpurineshasbeenassociatedwithseveralhealthdisorders,includinggoutandkidneystones.ThisstudyaimedtodeterminethepurinecontentinsoymilkusingtheUPLC(UltraPerformanceLiquidChromatography)methodandexploretheadsorptionofpurinesbyGranularCarbonBlack(GCB).
Introduction:
Soymilkisapopularalternativetodairymilk,especiallyamongindividualswithlactoseintoleranceorthosefollowingavegandiet.However,itisessentialtoassessthepurinecontentinsoymilkduetoitsassociationwithhealthissues.Previously,conventionalmethodssuchasHPLC(HighPerformanceLiquidChromatography)havebeenemployedforpurineanalysis,butthepresentstudyemployedUPLCduetoitshighefficiencyandimprovedresolution.
Methodology:
1.SamplePreparation:Severalsoymilksampleswereobtainedfromdifferentsourcesandbrandsandweresubjectedtocentrifugationfor10minutesat3000rpmtoremoveanysolidparticles.Thesupernatantwascollectedforanalysis.
2.UPLCAnalysis:AnAcquityUPLCsystemwithaWatersAcquityHSSC18columnwasusedfortheseparationandquantificationofpurines.Amobilephaseconsistingofamixtureof0.1%formicacidinwater(A)andacetonitrile(B)wasusedinagradientelutionmode.Thecolumntemperaturewassetat30°C,andtheflowratewasmaintainedat0.3mL/min.Thepurinecompoundsweredetectedat254nm.
ResultsandDiscussion:
TheUPLCanalysisrevealedthepresenceofseveralpurinecompoundsinsoymilk,includingadenine,guanine,hypoxanthine,andxanthine.Thecalibrationcurvesforeachpurinecompoundshowedgoodlinearity(R^2>0.99)withinthetestedconcentrationrange.Thepurinecontentindifferentsoymilksamplesvaried,withadeninebeingthemostabundantpurinecompoundpresent.ThetotalpurinecontentrangedfromXXtoXXmg/L.
Furthermore,theabilityofGCBtoadsorbpurineswasinvestigatedusingabatchadsorptionstudy.GCBwasaddedtoasoymilksample,andthemixturewasagitatedatroomtemperaturefordifferenttimedurations.Theconcentrat
