Abstract:
Pyrroloquinolinequinone(PQQ)isanessentialredoxcofactorwithavarietyofbiologicalfunctions.Inthisstudy,wereportthewholegenomesequencingandcomparativegenomicanalysisofanovelPQQ-producingmethylotrophicbacterium.Theanalysisofthegenomicdataprovidesvaluableinsightsintothegeneticcomposition,metabolicpotential,andevolutionaryrelationshipsofthisbacterium.

Introduction:
Pyrroloquinolinequinone(PQQ)isaredox-activecompoundthatplaysanimportantroleinvariousbiologicalprocesses,includingenergymetabolism,antioxidantactivity,andcellsignaling.PQQisproducedbycertainbacteria,fungi,andplants.Methylotrophicbacteriahaveattractedsignificantattentionfortheirabilitytoutilizeone-carboncompounds,suchasmethanol,methane,andformaldehyde,ascarbonandenergysources.SomemethylotrophicbacteriaareknowntoproducePQQ,makingtheminterestingcandidatesforstudyingPQQbiosynthesispathways.Inthisstudy,weaimtoinvestigatethegenomicfeaturesofanovelPQQ-producingmethylotrophicbacteriumthroughwholegenomesequencingandcomparativegenomicsanalysis.
Methods:
ThenovelPQQ-producingmethylotrophicbacteriumwasisolatedfromanenvironmentalsampleusingenrichmentculturetechniques.GenomicDNAwasextracted,andwholegenomesequencingwasperformedusinghigh-throughputsequencingtechnologies.Denovoassemblyandannotationofthegenomewereconductedfollowedbycomparativegenomicsanalysiswithrelatedspeciesusingavailablebioinformaticstools.ComparativegenomicsanalysisfocusedonidentifyingPQQbiosynthesisgenes,metabolicpathways,andpotentialhorizontalgenetransferevents.
Results:
ThewholegenomeofthenovelPQQ-producingmethylotrophicbacteriumwassequenced,andtheassemblyyieldedanapproximately4.5Mbgenome.ComparativegenomicsanalysisrevealedasetofgenesinvolvedinPQQbiosynthesis,includingpqqABCDEFGHIJ.Thesegenesshowed
