Abstract:
ThepresentstudyaimedtoestablishreliabledeterminationmethodsforthreesaponincomponentspresentinrattissuestreatedwithSanqiextract,aswellastoinvestigatethepharmacokineticsofthesecomponents.Inthisstudy,weutilizedhigh-performanceliquidchromatography(HPLC)coupledwithmassspectrometry(MS)toanalyzeandquantifythesaponincomponentsinrattissues.Thepharmacokineticpropertiesofthesecomponentswerethendeterminedandanalyzed.
Introduction:
Sanqiextract,derivedfromPanaxnotoginseng,isawidelyusedtraditionalChinesemedicineknownforitsmultifacetedpharmacologicaleffects.SaponinsarethemajorbioactivecomponentsofSanqiextractandarebelievedtocontributetoitstherapeuticeffects.However,thedeterminationmethodsforthesesaponincomponentsinbiologicalsamplesandtheirpharmacokineticpropertieshavenotbeenfullyelucidated.
Methods:
RatmodelswereestablishedbyorallyadministeringSanqiextract,andthebloodandtissuesampleswerecollectedatdifferenttimepoints.HPLC-MSwasemployedtoidentifyandquantifythesaponincomponentsintherattissues.Thepharmacokineticparameters,includingareaunderthecurve(AUC),peakplasmaconcentration(Cmax),andeliminationhalf-life(t1/2),werecalculatedusingnon-compartmentalanalysis.
Results:
Threesaponincomponents,namelyginsenosideRg1,Rb1,andnotoginsenosideR1,weresuccessfullyseparatedandidentifiedintherattissuestreatedwithSanqiextract.TheestablishedHPLC-MSmethodexhibitedgoodlinearity,precision,andaccuracy.Thepharmacokineticanalysisrevealedthatallthreesaponincomponentsshowedrapidabsorption,distribution,andeliminationfromtherattissues.Furthermore,ginsenosideRg1wasfoundtohavethehighestplasmaconcentrationandlongesthalf-lifeamongthethreecomponents.
Discussion:
TheestablishmentoftheHPLC-MSmethodallowedforaccurateandefficientdeterminationofthesaponincomponentsinrattissuestreatedwithSanqiextract.Theobtainedpharmacokineticparametersprovidedvaluableinsigh
