Abstract:
Fermentedyoghurtplaysasignificantroleinhumannutritionandhealthduetoitsprobioticproperties.Traditionalmethodsforbacterialanalysisinfermentedyoghurt,suchasplatecountingandPCR-basedtechniques,havelimitationsintermsofdetectionsensitivityandefficiency.Thisstudyaimstoinvestigatetheapplicationofsingle-moleculereal-time(SMRT)sequencingtechnologytoimprovethedetectionsensitivityofbacteriaintraditionalfermentedyoghurt.TheresultsdemonstratethatSMRTsequencingenablestheidentificationandquantificationofbacteriainyoghurtsampleswithunparalleledresolutionandaccuracy.ThesefindingssuggestthattheintegrationofSMRTsequencingtechnologyintotheanalysisoffermentedyoghurtcangreatlyenhanceourunderstandingofthemicrobialcompositionandprovidevaluableinsightsintothefermentationprocess.
Introduction:
Fermentedyoghurtisapopulardairyproductconsumedworldwideandisrecognizedforitshealthbenefitsattributedtothepresenceoflivemicroorganisms,particularlylacticacidbacteria(LAB)andbifidobacteria.Traditionalmethodsofbacterialanalysisinyoghurt,suchasplatecountingandPCR-basedtechniques,havelimitationsintermsofsensitivity,accuracy,andefficiency.Theselimitationshinderourabilitytofullyunderstandthecomplexmicrobialdynamicsandcompositionoffermentedyoghurt.Thus,thereisaneedforamoreadvancedandhigh-throughputmethodtoanalyzebacterialpopulationsinyoghurtsamples.
Single-moleculereal-time(SMRT)sequencingtechnology,developedbyPacificBiosciences,offersaninnovativeapproachforDNAsequencing.Thistechnologyproduceslongreadswithhighaccuracyandenablesreal-timemonitoringofsingleDNAmoleculesduringthesequencingprocess.SMRTsequencinghasbeensuccessfullyappliedinvariousfields,includingthehumanmicrobiome,pathogenidentification,andmetagenomics.However,itsapplicationintheanalysisoffermentedyoghurthasnotbeenextensivelyexplored.
