Abstract:
Surface-enhancedRamanscattering(SERS)hasemergedasapowerfulanalyticaltechniquefordetectingtraceanalytes,includingmicroRNAs(miRNAs),duetoitshighsensitivityandselectivity.Inthisstudy,wepresentanovelthree-dimensionalSERSsubstratebasedonananopillarforestformiRNAdetection.Thesubstratewasfabricatedusingacombinationofnanoimprintlithographyandmetaldepositiontechniques.TheSERSenhancementpropertiesofthesubstratewereoptimizedthroughsystematicinvestigationofthesize,shape,anddensityofthenanopillars.Inaddition,themiRNAdetectionperformanceoftheSERSsubstratewasevaluatedbyimmobilizingcomplementaryDNAprobesontothenanopillarsurface.Theresultsdemonstratethatthedevelopedthree-dimensionalSERSsubstrateoffershighsensitivityandexcellentreproducibilityforthedetectionofmiRNA.
1.Introduction:
MicroRNAs(miRNAs)areshortnon-codingRNAsthatplaycrucialrolesintheregulationofgeneexpression.AbnormalexpressionlevelsofspecificmiRNAshavebeencorrelatedwithvariousdiseases,includingcancer,cardiovasculardiseases,andneurodegenerativedisorders.Therefore,thedevelopmentofsensitiveandreliablemiRNAdetectionmethodsisofgreatimportanceforbothbasicresearchandclinicaldiagnostics.Surface-enhancedRamanscattering(SERS)hasdemonstratedoutstandingcapabilitiesformiRNAdetectionduetoitsultrasensitivityandmolecularspecificity.However,traditionalplanarSERSsubstrateshavelimitationsintermsofreproducibilityandsensitivity.Theintroductionofthree-dimensionalSERSsubstrates,suchasnanopillarforests,hasshowngreatpromiseinovercomingtheselimitations.
2.FabricationofThree-DimensionalSERSSubstrate:
Thefabricationofthethree-dimensionalSERSsubstrateinvolvedatwo-stepprocess.First,ananopillarforestwaspreparedbynanoimprintlithography,whichenabledprecisecontroloverthesize,shape,anddensityofthenanopillars.Then,athinmetalliclayer(e.g.,goldorsilver)wasdepositedontothenanopillarsurface,providingSERSenhancement.Thethicknessofthemetallayerwasop
