HPLCfingerprintanalysisandidentificationofSourJujubeKernelandYunnanJujubeKernel
Introduction
SourJujubeKernel(SJ)andYunnanJujubeKernel(YJ)aretwocommonlyusedtraditionalChinesemedicineswithsimilarnamesandappearances.Theyareoftenusedinterchangeably,buttheyhavedifferenttherapeuticeffectsandchemicalcompositions.Inordertoensuretheirclinicalsafetyandefficacy,itisnecessarytoestablishareliablemethodfortheiridentificationandqualitycontrol.Highperformanceliquidchromatography(HPLC)fingerprintanalysisisapowerfulandwidelyacceptedmethodforthecharacterizationandidentificationofherbalmedicines.Inthisstudy,wedevelopedandvalidatedanHPLCfingerprintmethodfortheanalysisandidentificationofSJandYJ.
MaterialsandMethods
Materials:
SJandYJwerepurchasedfromareputableChinesemedicinesupplier.
MethanolandacetonitrileHPLC-gradewerepurchasedfromSigma-Aldrich.
DeionizedwaterwaspreparedusingaMilliporepurificationsystem.
Instrumentation:
HPLCanalyseswereperformedonanAgilent1200seriessystemequippedwithaphotodiodearraydetector(DAD).ChromatographicseparationswerecarriedoutonaC18column(250mm×4.6mm,5µmparticlesize)atacolumntemperatureof30°C.
Mobilephase:
Themobilephaseconsistedofacetonitrile(A)andwatercontaining0.1%phosphoricacid(B),usingagradientelutionsystemasfollows:0-25min,10-38%A;25-35min,38-48%A;35-50min,48-75%A;50-65min,75-100%A;65-70min,100-10%A.
Samplepreparation:
SJandYJweregroundintofinepowdersandaccuratelyweighed(1.0g)andextractedwith50mLofmethanolinanultrasonicbathfor30min.Theextractswerefilteredthrougha0.22µmnylonfilterbeforeHPLCanalysis.
Methodvalidation:
TheHPLCfingerprintmethodwasvalidatedforitsprecision,stability,repeatability,accuracy,androbustness.Theprecisionwasevaluatedbyinjectingsixreplicatesofthesamesamplesolution.Thestabilitywasevaluatedbyanalyzingthesamesamplesolutionat0,2,4,6,8,12,and24hafterpreparation.Therepeatabilitywasevaluatedbyanalyzingsixdifferentsamplesfromthesamebatch.TheaccuracywasevaluatedbyspikingknownamountsofSJorYJintotheirrespectivesamples
