Introduction
ThPOK(T-helperinducingPOZ/Krüppel-likefactor)isatranscriptionfactorthatplaysacrucialroleinthedifferentiationandmaintenanceofCD4+Thelper(Th)cells.Itisknowntoberegulatedbymultiplesignalingpathwaysandpost-translationalmodifications,includingacetylation.Inthispaper,wewillfocusontheacetylationofThPOKanditsroleindirectingCD4+CD8+Tcelldifferentiation.
AcetylationofThPOK
Acetylationisapost-translationalmodificationthatinvolvestheadditionofanacetylgrouptoalysineresidueonaprotein.Thismodificationisimportantintheregulationoftranscriptionfactoractivity.ThPOKhasbeenshowntobeacetylatedbythehistoneacetyltransferase(HAT)p300/CBP,whichisknowntoregulatetheacetylationofmanytranscriptionfactors.

ThPOKplaysacriticalroleinthedifferentiationofCD4+Tcells.IthasbeenshowntorepresstheexpressionofgenesthatpromoteCD8+TcelldifferentiationandtopromotetheexpressionofgenesthatpromoteCD4+Tcelldifferentiation.ThisfunctionisparticularlyimportantinthedifferentiationofCD4+CD8+Tcells,whichhavethepotentialtodifferentiateintoeitherCD4+orCD8+Tcells.
RecentstudieshaveshownthattheacetylationofThPOKisimportantindirectingCD4+CD8+TcelldifferentiationtowardstheCD4+Tcelllineage.Specifically,acetylationofThPOKatK252hasbeenshowntopromotethedifferentiationofCD4+TcellsandtoinhibitthedifferentiationofCD8+Tcells.
ThemechanismbywhichacetylationofThPOKpromotesCD4+Tcelldifferentiationisnotyetfullyunderstood.However,ithasbeensuggestedthatacetylationofThPOKmayenhanceitsinteractionwithco-activators,suchasp300/CBP,leadingtoincreasedtranscriptionofgenesthatpromoteCD4+Tcelldifferentiation.
