[Abstract]
Embryonicstemcellsareaspecialtypeofstemcellsthathavethepotentialtodifferentiateintoanycelltypeinthebody.NanogandOCT4aretwokeytranscriptionfactorsthatplayimportantrolesinmaintainingthepluripotencyofembryonicstemcells.ThisreviewfocusesonrecentstudiesonthefunctionsofNanogandOCT4inembryonicstemcells,includingtheirrolesinpluripotencymaintenance,cellularreprogramming,andcellulardifferentiation.
[Introduction]
Embryonicstemcells(ESCs)arederivedfromtheinnercellmassoftheblastocystandhavethepotentialtodifferentiateintoanycelltypeinthebody.ThepluripotencyofESCsismaintainedbyacomplexnetworkoftranscriptionfactors,includingNanogandOCT4.Nanogwasfirstidentifiedasafactorthatmaintainsself-renewalandpluripotencyinmouseESCs(mESCs)in2003,whileOCT4wasfoundtobeessentialformaintainingthepluripotencyofbothmouseandhumanESCs(hESCs)intheearly2000s.Sincethen,numerousstudieshavebeenconductedtoinvestigatetherolesofNanogandOCT4inESCs.
[Pluripotencymaintenance]
NanogandOCT4aretwokeytranscriptionfactorsthatarecrucialformaintainingthepluripotencyofESCs.Theyworktogetherwithothertranscriptionfactors,suchasSOX2,toformaregulatorynetworkthatcontrolsgeneexpressionandmaintainspluripotency.Nanoghasbeenshowntobeessentialfortheself-renewalandpluripotencyofmESCs,aslossofNanogexpressionleadstodifferentiationofmESCsintoextraembryonicendodermortrophectoderm.Similarly,OCT4isalsorequiredforthemaintenanceofpluripotency,aslossofOCT4expressionleadstodifferentiationofESCsintoprimitiveendoderm.TheexpressionofNanogandOCT4istightlyregulatedduringearlyembryonicdevelopment,andalterationsintheirexpressionlevelscandisruptthebalancebetweenself-renewalanddifferentiationofESCs.
[Cellularreprogramming]
NanogandOCT4havealsobeenshowntoplayimportantrolesincellularreprogramming.Cellularreprogrammingistheconversionofadifferentiatedcellintoapluripotentormultipotentstemcellorintoadifferentcelltypeusingacombinationoftranscriptionfactorsorothermolecules.Thegenerationofinducedpluripotentstemcells(iPSCs)fromso
