Abstract:
Theshortlateralbranchingtraitinmelon(Cucumismelo)isanimportantagronomictraitthataffectsfruityieldandplantarchitecture.Inthisstudy,weperformedQTLmappingtoidentifythegeneticlociresponsiblefortheshortlateralbranchingtraitinmelon.Amappingpopulationwascreatedbycrossingtwomelonvarieties,onewithashortlateralbranchingphenotypeandtheotherwithanormalbranchingphenotype.TheF1progenywereselfedtogenerateanF2populationwhichwasthenphenotypedfortheshortlateralbranchingtrait.Genotypingwasperformedusingmolecularmarkers,andQTLanalysiswasconductedtoidentifythegenomicregionsassociatedwiththetrait.
Introduction:
Melonisaneconomicallyimportantfruitcropworldwide,andplantarchitectureplaysacrucialroleindeterminingfruityieldandquality.Theshortlateralbranchingtrait,characterizedbyreducedbranchingonthemainstem,hasbeenreportedtoincreasefruityieldbyconcentratingresourcesonfruitdevelopment.Therefore,understandingthegeneticbasisofthistraitisofgreatsignificanceformelonbreedingprogramsaimedatimprovingyieldandplantarchitecture.
Methods:
1.Plantmaterials:Twomelonlines,onewithshortlateralbranchingandtheotherwithnormalbranching,wereselectedasparentallinesforthecross.
2.Populationdevelopment:TheparentallineswerecrossedtoobtainF1plants,whichwereselfedtogenerateanF2population.
3.Phenotyping:TheF2populationwasphenotypedfortheshortlateralbranchingtraitbyvisuallyassessingthenumberandlengthoflateralbranchesonthemainstem.
4.Genotyping:DNAwasextractedfromtheF2population,andasetofmolecularmarkers,suchasSSRsorSNPs,wereusedtogenotypethepopulation.
5.QTLmapping:QTLanalysiswasperformedusingappropriatestatisticalsoftware,suchasQTLCartographerorMapQTL,toidentifythegenomicregionsassociatedwiththeshortlateralbranchingtrait.
6.Validation:TheidentifiedQTLswerefurthervalidatedusingadditionalmolecularmarkersandbyanalyzingtheireffectsontheshortlateralbranchingtrait.
Results:
Atotalof200F2plantswerephenotypedfortheshortlateralbranchingtrait
